Assay Principles and Components

Protease assay kits typically rely on protease-mediated cleavage of a protein substrate, which releases detectable products measured either colorimetrically or fluorometrically.

Colorimetric Assays: A common approach uses succinylated casein as a substrate, which is cleaved by proteases to expose primary amines. These amines react with 2,4,6-trinitrobenzenesulfonic acid (TNBSA) to produce an orange-yellow chromophore measured at 450 nm. The increase in absorbance correlates with protease activity. This method is homogeneous, performed in microplates without requiring separation steps, and uses trypsin as a standard for calibration.

Fluorometric Assays: Fluorescently labeled casein substrates (e.g., fluorescein-conjugated casein) are cleaved by proteases, releasing fluorescent peptides. The fluorescence intensity, measured at excitation/emission wavelengths such as 490/525 nm, increases proportionally with protease activity. This method offers higher sensitivity and can detect lower protease concentrations compared to colorimetric assays.

Sensitivity and Specificity

Protease assay kits can detect a broad spectrum of proteases, including serine proteases (trypsin, chymotrypsin), cysteine proteases (papain), and acid proteases (pepsin). Sensitivity varies by assay type; fluorometric assays generally offer greater sensitivity, capable of detecting nanogram levels of protease. Colorimetric assays are simpler and faster.
