Proteome Profiler Cell Stress Array Kit, BioAssay™ (Human)

The Proteome Profiler Human Cell Stress Array Kit is a membrane-based sandwich immunoassay. Samples are mixed with a cocktail of biotinylated detection antibodies (Step 1) and then incubated with the array membrane which is spotted in duplicate with capture antibodies to specific target proteins (Step 2). Captured proteins are visualized using chemiluminescent detection reagents (Step 3). The signal produced is proportional to the amount of analyte bound.

Principle of the Assay:
Capture and control antibodies have been spotted in duplicate on nitrocellulose membranes. Cell lysates or tissue lysates are diluted, mixed with a cocktail of biotinylated detection antibodies, and incubated overnight with the Proteome Profiler Human Cell Stress Array. The membrane is washed to remove unbound material. Streptavidin-HRP and chemiluminescent detection reagents are applied, and a signal is produced at each capture spot corresponding to the amount of protein bound. Kit Components:
Human Cell Stress Array: 4 nitrocellulose membranes each containing
26 different capture antibodies printed in duplicate.
Array Buffer 4, 21ml of a buffered protein base with preservatives. May contain a precipitate.
Array Buffer 6: 2 vials (21ml/vial) of a buffered protein base with preservatives. May contain a precipitate.
Lysis Buffer 6: 21ml of a denaturing buffered solution.
Wash Buffer Concentrate: 2 vials (21ml/vial) of a 25-fold concentrated solution of buffered surfactant with preservative. May turn yellow over time.
Detection Antibody Cocktail, Human Cell Stress Array: 1 vial of biotinylated antibody cocktail; lyophilized.
Streptavidin-HRP: 200ul of streptavidin conjugated to horseradish-peroxidase.
Chemi Reagent 1: 1 vial (2.5ml)
Chemi Reagent 2: 1 vial (2.5ml)
4-Well Multi-dish: Clear 4-well rectangular multi-dish.
Transparency Overlay Template: 1 transparency overlay template for coordinate reference.

Storage and Stability:
Unused kit is stable for 3 months at 4°C. Once opened may be stored for up to 3 months at 4°C; return unused membranes to the foil pouch containing the dessicant pack and reseal along entire edge of the zip seal.
