Description

IGF signaling (putative; PI3K/AKT, MAPK); roles in growth and metabolic regulation (putative). Background of IGF1

Insulin-like growth factor 1 (IGF-1), also called somatomedin C, is a protein that in humans is encoded by the IGF1 gene. It is mapped to 12q23.2. The protein encoded by this gene is similar to insulin in function and structure and is a member of a family of proteins involved in mediating growth and development. The encoded protein is processed from a precursor, bound by a specific receptor, and secreted. Defects in this gene are a cause of insulin-like growth factor I deficiency. Alternative splicing results in multiple transcript variants encoding different isoforms that may undergo similar processing to generate mature protein.

The EK0376 Human IGF-1 ELISA Kit is designed to measure free IGF1. IGF-1 is usually bound to IGFBPs in the serum/plasma, so if you would like to measure total IGF1, it is advisable to release the analytes by performing an extraction procedure such as acid-ethanol treatment.

I am interested purchasing EK0376 Human IGF-1 ELISA Kit for serum or plasma samples. I read the protocol calls for pre-treatment of samples to separate IGF1 from the IGF1 Binding Proteins in order to accurately measure total IGF1. Is it possible not performing the extraction step and only measure free IGF1 (not initially bound to IGF1BPs) in serum or plasma? ? or have you ever tried it?

Hello; the purpose of the extraction step is to increase the signal, as the antibody EK0376 Human IGF-1 ELISA Kit recognition is apparently blocked by the binding protein interfering with the epitopes that one or both of the antibodies bind to. Probably some signal will be obtained even without the extraction step, but the EK0376 Human IGF-1 ELISA Kit will not be able to differentiate signal contributed by free EGF1 from the signal contributed by EGF1 in complex with binding protein.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may change the stabilityof samples. It is recommend to assay sample on-stat collection when possible, or aliquot into single use volumes and store samples frozen. avoid repeat freeze-thaw cycles with the stored samples to limit protein degradation.

Q: can you recommend the dilution ratio of serum samples for detection of IGF-I in Human plasma? I am trying to measure a few analytes and it requires 100ul of diluted samples for each well. We have limited sample volumes so we like to dilute as much as possible.

A: without having an understanding the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the IGF-I ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.
