HigherPurity™ Yeast DNA Isolation Kit

Efficient kit for high-quality genomic DNA purification from yeast.

The HigherPurity™ Yeast DNA Isolation Kit provides a fast, consistent, and efficient method for purifying genomic DNA from yeast and other fungal samples. It combines lyticase enzymatic digestion, glass bead disruption, and alkaline SDS lysis, followed by silica column binding to ensure inhibitor-free DNA recovery.

Purified DNA is suitable for PCR, sequencing, Southern blotting, and restriction enzyme digestion. The complete workflow takes under 90 minutes and consistently delivers high-quality DNA free of contaminants.

For filamentous fungi or macroscopic fungi, the yeast protocol may be used as a reference and may require optimization.

Quality Control

  • Tested lot-to-lot by isolating DNA from 50 mg yeast.

  • DNA quality confirmed by spectrophotometer (A260/A280 = 1.7–1.9).

  • Integrity checked by agarose gel electrophoresis.

Advice

  • Store Proteinase K at –20 ºC for long-term stability.

  • Ensure efficient homogenization with glass beads for optimal yields.

Storage, Shipping & Guarantee

  • Shipped in: Ambient Temperature.
  • Storage*: All components can be stored at Room Temperature, except RNAse A, Rupture Buffer  and Proteinase K (4 ºC). Proteinase K should be stored at -20 ºC for longer-term storage.

FAQs

Q: How long does the protocol take?
A: Around 88 minutes.

Q: Is phenol/chloroform extraction required?
A: No, the protocol is safe and chemical-free.

Q: How can DNA yield be improved when working with yeast or mould samples?
A: Yield can be improved by ensuring complete spheroplast formation (adequate lyticase incubation) and efficient mechanical disruption during bead beating.

Q: Can lyticase incubation time be extended to improve lysis efficiency?
A: Yes, incubation can be extended from 30 minutes up to 45–60 minutes at 30 °C to improve spheroplast formation in difficult samples.

Q: Is it possible to adjust the lysis buffer volume for dense samples?
A: A slight increase in Buffer BLY volume (10–20%) can improve homogenization of dense pellets.
