Carlson Lysis Buffer is used in DNA purification of plant, bacteria, and fungal sources. CTAB lyses cell membranes to make the DNA accessible for extraction, while efficiently removing both polysaccharides and residual proteins.

This buffer is prepared using raw materials only of >99% purity and higher, and is of Molecular Biology Grade to be used directly in DNA extraction.

For best results, add 20 mL of Carlson Lysis Buffer to 1-2 g of powdered sample. Add 50 µL of beta-Mercaptoethanol and 200 µL of RNase A (provided at 20 mg/mL) and incubate for 20 minutes at 74°C. When the system is at ambient temperature, add Chloroform/ Isoamyl Alcohol 24:1 and vortex. After centrifuging, use Hydrochloric Acid to pH the supernatant to 7.0.
