This biotinylated antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 50 µg of bovine serum albumin per µg of antibody with no calcium, magnesium, or preservatives present.

The lyophilized, biotinylated antigen affinity purified polyclonal antibody can be stored desiccated at 20°C to 70°C for up to twelve months from date of receipt. The reconstituted biotin conjugate can be stored for at least four weeks at 28°C. For longterm storage of the reconstituted conjugate, aseptically aliquot into working volumes and store at 20°C to 70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Rat IL1α this biotin conjugate can be used at a concentration of 0.1 µg/ml. This biotin conjugate should be used in conjunction with compatible secondstep reagents such as PN:A106 and a chromogenic substrate such as PN:T343. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:I334.
ELISA Sandwich Assay: This antibody can be used as the detection antibody in a sandwich ELISA at a concentration of approximately 0.10.4 µg/ml when used in conjunction with PN:I671 as the capture antibody at 28 µg/ml and an optimal second step reagent such as PN:A106 for the detection of Rat IL1α.

Goat AntiRat Interleukin1α (IL1α) recognizes Rat IL1α. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background
IL1R1 is an 80 kD transmembrane glycoprotein and a member of the Ig superfamily. IL1 binds both IL1R1 and IL1R2. The ligands IL1α and IL1β only commence when binding IL1R1 because IL1R2 acts as a decoy receptor and competitively antagonizes the binding of IL1α and IL1β, thereby reducing their biological effects. Binding of the ligands to IL1R1 is enhanced by an accessory protein, IL1RAcP. IL1α can exist as either a 31 kD precursor or an 18 kD mature form. IL1α is a proinflammatory cellassociated cytokine and IL1β is a 17.5 kD proinflammatory secreted cytokine. Both IL1α and IL1β bind to the same receptor and have similar properties that include stimulation of thymocyte proliferation via IL2 release, Bcell maturation and proliferation, mitogenic FGFlike activity and the ability to stimulate the release of prostaglandin and collagenase from synovial cells. Furthermore, IL1β mediates a variety of immune and inflammatory responses. IL1β is a major cause of severe inflammation in the mouse model of Crohn’s Disease, which makes IL1β an important target for this disease.
