Applications:
Suitable for use in Western Blot, Immunocytochemistry, Immunoprecipitation and ELISA. Other applications not tested.
Recommended Dilution:
Western Blot: 0.5-2ug/ml detects cdk5 in a RIPA lysate from non-stimulated human A431 cells. Non-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C2575-01 (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Previous lots of antibody detected cdk5 in RIPA lysates from mouse Wehi cells and rat PC12 cells.
Immunoprecipitation: 4ug/500ug of A431 cell lysate.
Immunocytochemistry: 5ug/ml stains cdk5 in human A431 cells fixed with 4% paraformaldehyde.
Optimal dilutions to be determined by the researcher.
Included Positive Antigen Control:
A0001-50 A431 Cells, Non-Stimulated. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.

Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
