Inulin

Inulin is a naturally occurring plant-derived fructan and non-digestible storage polysaccharide classified as a prebiotic dietary fiber. Primarily extracted from chicory roots, it is valued for its health-promoting effects on gut microbiota.

Molecular Structure

Inulin consists of linear chains of β-(2→1)-linked D-fructofuranose units, typically terminated at the reducing end by a single α-D-glucopyranosyl residue. Branching is minimal (≤5%) via β-(2→6) linkages. The degree of polymerization (DP) of standard inulin ranges from 2-60, with an average DP of 10-12. High-performance inulin variants, produced by removing short-chain fructooligosaccharides (DP<10, such as 1-kestose), exceed DP 23. Molecular weights span 1-5 kDa, and nuclear magnetic resonance (NMR) spectra show characteristic anomeric signals at δ 4.2-5.4 ppm.

Extraction and Properties

Inulin extraction typically involves hot water extraction (70-90°C) from Cichorium intybus roots, solubilizing inulin with yields of approximately 10-20% of dry weight. The extract is clarified with activated carbon, purified via ion-exchange, and dried by spray-drying. Enzymatic hydrolysis can produce shorter oligofructose fractions. Inulin exhibits high water solubility (>100 g/L at 25°C), low viscosity, thermal stability (gelatinization above 140°C), non-hygroscopic behavior, and a low glass transition temperature due to its flexible fructosyl backbone. The crystalline powder form self-assembles into helical structures stabilized by hydrogen bonding.

Biomedical Applications

As a prebiotic, inulin is selectively fermented by beneficial gut bacteria such as Bifidobacteria and Lactobacilli. Fermentation produces short-chain fatty acids (SCFAs), which contribute to gut barrier integrity, immunomodulation, and reduced inflammation. Inulin intake is associated with a lower glycemic index, improved calcium absorption, and support for weight management. In pharmaceutical contexts, high-DP inulin is explored for protein stabilization, colon-targeted nanoparticle drug delivery, and potential anticancer effects via the induction of apoptosis in tumor cells.
