Anti-TFE3 CE/IVD antibodies are specialized immunohistochemistry reagents developed for the detection of Transcription Factor E3 (TFE3), a member of the microphthalmia-associated transcription factor (MiT/TFE) family of basic helix-loop-helix leucine zipper transcription factors. TFE3 regulates genes involved in cellular metabolism, lysosomal biogenesis, autophagy, cell growth, differentiation, and immune signaling through binding to E-box DNA sequences and interaction with multiple transcriptional pathways. TFE3 is constitutively expressed in many normal tissues, typically at low to moderate levels, whereas gene rearrangements frequently result in marked nuclear accumulation detectable by immunohistochemistry.

Biological Significance of TFE3

  • Nuclear transcription factor belonging to the MiT/TFE family together with TFEB, TFEC, and MITF.
  • Regulates cellular proliferation, metabolic adaptation, autophagy, lysosomal biogenesis, and cellular homeostasis.
  • Oncogenic activation most frequently results from chromosomal translocations generating TFE3 fusion proteins with aberrant transcriptional activity and dysregulated gene expression.

Diagnostic Utility of TFE3 in Pediatric Pathology

  • Key diagnostic marker for alveolar soft part sarcoma (ASPS), a tumor frequently affecting children and young adults and characterized by the ASPSCR1–TFE3 fusion.
  • Useful in the identification of Xp11 translocation renal cell carcinoma, an uncommon but clinically important pediatric renal neoplasm driven by TFE3 rearrangements.
  • May support diagnosis of rare TFE3-rearranged PEComas and other MiT family translocation-associated tumors when interpreted within an immunohistochemical panel and correlated with molecular findings.
  • Diffuse strong nuclear staining raises suspicion for TFE3 rearrangement; however, confirmatory molecular testing such as FISH, RT-PCR, or RNA sequencing remains recommended because antibody performance and specificity may vary among clones, tumor types, and preanalytical conditions.
