Anti-Human Germinal Center–Associated Lymphoma (HGAL) CE/IVD antibodies are essential IHC tools in hematopathology for identifying germinal center (GC) B-cell origin and refining lymphoma classification. HGAL (also known as GCET2) is a GC-associated protein discovered through gene-expression profiling of B-cell lymphomas and is recognized as a useful immunohistochemical marker in diagnostic pathology.

Biological Significance of HGAL

HGAL is a germinal center–associated protein expressed predominantly in normal GC B lymphocytes and GC-derived lymphomas.

  • Expressed mainly in GC B cells and typically not detected in most non–germinal center lymphoid compartments.
  • Involved in the regulation of lymphocyte motility through pathways including RhoA signaling, contributing to controlled cellular migration.
  • Expression has been associated with more favorable outcomes in certain lymphoma studies, including diffuse large B-cell lymphoma (DLBCL), although it is not used as a standalone prognostic marker in routine practice.
  • Identified as part of germinal center transcriptional signatures derived from molecular profiling studies.

Diagnostic Utility in Hematopathology

HGAL is a useful immunohistochemical biomarker for supporting germinal center derivation and is commonly used in combination with CD10 and BCL6.

  • Frequently expressed in follicular lymphoma, Burkitt lymphoma, and germinal center B-cell–type DLBCL.
  • Provides complementary information alongside established GC markers such as CD10 and BCL6.
  • Helps support distinction between germinal center (GC) and non–germinal center (non-GC) DLBCL subtypes in conjunction with other markers.
  • Assists in identifying germinal center–derived B-cell populations in disrupted lymphoid architecture and atypical lymphoid proliferations.
  • May show expression in a subset of Hodgkin lymphoma cases, though its diagnostic utility in this context is limited and not routine.
