Fruit allergens

Fruit allergens

Fruit allergens are a key focus in molecular allergy research, biotechnology, and component-resolved diagnostics (CRD), enabling precise characterization of IgE-mediated hypersensitivity and pollen–food allergy syndrome. Defined allergen molecules are widely used in recombinant protein production, immunoassay development, and multiplex diagnostic platforms to support both clinical and translational research applications.

Among well-characterized fruit allergens, Mal d 1 (Malus domestica, apple) is a PR-10 protein homologous to birch pollen Bet v 1, widely associated with pollen–food cross-reactivity and typically linked to mild, heat-labile reactions. In strawberry (Fragaria × ananassa), Fra a 1 (PR-10) and Fra a 4 (profilin) are identified IgE-binding components involved in cross-reactive sensitization profiles, though their clinical relevance is generally variable and population-dependent.

In mango (Mangifera indica), identified IgE-reactive components include profilins and chitinase-related proteins, but allergen nomenclature and clinical validation remain less standardized compared with major Rosaceae fruit allergens, reflecting ongoing research in tropical fruit allergen profiling.

Key Fruit Allergen Families Relevant for Biotechnology and CRD

  • PR-10 proteins (e.g., Mal d 1, Fra a 1): labile, cross-reactive allergens used in pollen–food syndrome studies.
  • Profilins (e.g., Fra a 4): highly conserved pan-allergens enabling broad IgE cross-reactivity analysis.
  • Lipid transfer proteins (LTPs): stable allergens associated with severe systemic reactions, particularly in Rosaceae fruits.
  • Defense-related proteins: including chitinase-associated allergens in selected species investigated for structural stability and IgE binding properties.

Biotechnological Applications

  • Recombinant allergen production (bacterial and yeast systems).
  • Purified native allergen isolation for analytical and diagnostic use.
  • ELISA and basophil activation assays for IgE binding profiling.
  • Multiplex CRD platforms for component-resolved allergy diagnostics.
  • Epitope mapping and allergen standardization workflows.
