B-cell lymphoma-6 (BCL6) is a nuclear transcriptional repressor encoded by the BCL6 proto-oncogene and is widely used as an immunohistochemical marker in hematopathology. Anti-Bcl-6 CE/IVD antibodies are routinely applied to detect BCL6 protein expression in formalin-fixed, paraffin-embedded (FFPE) tissue sections, supporting the classification of B-cell lymphomas and germinal-center–derived malignancies.

Biological Significance of Bcl-6

  • Master regulator of germinal center (GC) B cells: BCL6 is essential for germinal center formation and regulates the transcriptional program that enables rapid proliferation and antibody affinity maturation.
  • Transcriptional repression network: BCL6 binds numerous promoters and represses genes involved in DNA-damage responses, cell-cycle control, and differentiation, allowing GC B cells to tolerate somatic hypermutation and clonal expansion.
  • Oncogenic role in lymphomagenesis: Deregulated or constitutive BCL6 expression contributes to the development and maintenance of several B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL).

Diagnostic Utility of Bcl-6 in Hematopathology

  • Germinal-center B-cell marker: BCL6 protein is physiologically expressed in centroblasts and centrocytes within lymphoid follicles.
  • Differential diagnosis of lymphoid neoplasms: BCL6 immunostaining helps identify germinal-center–derived lymphomas such as follicular lymphoma, diffuse large B-cell lymphoma, and Burkitt lymphoma.
  • IHC classification algorithms: In DLBCL, BCL6 expression is integrated with markers such as CD10 and MUM1 to distinguish germinal-center B-cell (GCB) and activated B-cell (ABC) subtypes.
