4°C Do Not FreezeMast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. Human mast cell tryptase has considerable potential as a marker for mast cell activation as well as being a mediator of inflammation.
Applications:
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions:
Immunohistochemistry: 1:100-1:500. Tissues or cytospin preparations should be fixed in Carnoy’s Fluid (60% ethanol, 30% chloroform, 10% glacial acetic acid). Not recommended for use on formaldehyde fixed tissue. May be used on thin sections prepared for ultrastructural analysis after fixation of tissue in a modified Karnovsky’s reagent.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Store product at 4°C. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
