The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at 20°C to 70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 28°C. For longterm storage of the reconstituted antibody, aseptically aliquot into working volumes and store at 20°C to 70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human IL12 this polyclonal antibody can be used at a concentration of 0.1 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible secondstep reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Human IL12 is 5 ng/lane under either reducing or nonreducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.

Neutralization: This antibody is useful for neutralization of Human IL12 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human IL12 (at 1.0 ng/ml) is 0.03 0.05 µg/ml.

Goat AntiHuman Interleukin12 (IL12) recognizes Human IL12. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background
IL12 plays a role in resistance against pathogens via the differentiation of naive T cells into Th1 cells. It stimulates the growth and function of T cells, blocks formation of new blood vessels, and contributes to antimycobacterial immune response. In addition, it promotes the production of IFNγ and TNFα and reduces IL4 mediated suppression of IFNγ. Consequently, this enhances the immunostimulatory and immunomodulatory effects of IFNγ. In addition, there appears to be a link between IL2 and the signal transduction of IL12 in NK cells, which enhances the functional response of IL12 via IFNγ production and killing of target cells. Furthermore, IL12 is thought to be associated with autoimmunity. IL12 was shown to worsen the condition when administered to people already suffering from autoimmune diseases. Comparatively, inhibition of IL12 (either through IL12 gene knockout in mice or treatment of mice with IL12 mAbs) improved the disease.
