Anti-ChAT (Choline Acetyltransferase) Antibody

Our goat ChAT (Choline Acetyltransferase) polyclonal antibody from PhosphoSolutions is produced in-house. It detects avian, chicken, guinea pig, human, mouse, non-human primate, opossum, rat, and zebrafish ChAT (Choline Acetyltransferase) and is affinity purified. Works in ELISA, WB, IHC.

Western blot detection of ChAT using Anti-Choline Acetyltransferase (ChAT) Antibody (Cat. 315-CHAT). Gel was loaded with Mouse brain lysate. Proteins were transferred to the PVDF membrane, blocked, and then washed with a buffer. Primary antibody was incubated with sample at 1:1000 and then detected via chemiluminescence using rabbit anti-goat secondary antibody conjugated to HRP. Arrow indicates protein ChAT (~68-70 kDa).

Scientific Background

Choline acetyltransferase is a neuronal enzyme which catalyzes the reaction between Acetyl CoA and choline resulting in the formation of acetylcholine. It is therefore found primarily in cholinergic neurons making it a valuable marker for diseases associated with decreased cholinergic function such as Schizophrenia, Alzheimer's disease (AD) and Down syndrome (Holt et al. 1999). Decreased choline acetyltransferase activity in particular has been shown in Schizophrenic subjects (Karson et al 1993). It has furthermore been demonstrated that in patients with AD, there are significantly lower levels of cortical ChAT that correlate with severity of the disease as measured by loss of neuropsychological function (Baskin et al. 1999).

Recommended that the undiluted antibody be aliquoted into smaller working volumes (10-30 uL/vial depending on usage) upon arrival and stored long term at -20° C or -80° C, while keeping a working aliquot stored at 4° C for short term. Avoid freeze/thaw cycles. Stable for at least 1 year.

Application Details

Validated for use in ICC, IHC(P) and WB. Immunogen: human placental enzyme. IHC: 1:100 (previous lot). Controls: human placenta lysates and rat forebrain/rostral hypothalamus. Routinely evaluated by WB on mouse brain lysates (1:1000 detected CHAT on 10 ug mouse brain lysate). Optimal working dilutions and protocols must be determined by end user.
